The diets and feeding guild structure of a freshwater reed belt fish assemblage (30 species) were examined in Lake Kitaura, part of the Lake Kasumigaura system, eastern Japan, from June to September in 2009 and 2010. Ontogenetic dietary shifts were recognized in 14 species, including several species (e.g., Cyprinus carpio, Tridentiger brevispinis and Hyporhamphus intermedius) targeted by local fisheries. Juveniles of these species generally fed on zooplankton, later switching to other prey items (e.g., benthic chironomid larvae, gammaridean amphipods, shrimps, juvenile fishes, diatoms and decomposing reeds) with growth. A cluster analysis based on dietary overlaps showed that the reed belt fish assemblage comprised five feeding guilds (plant, zooplankton, benthic invertebrate, terrestrial insect and fish feeders). Of these, zooplankton feeders were the most abundantly represented in terms of species numbers, suggesting that the main food items for the fish assemblage were zooplankton (e.g., cladocerans, and calanoid and cyclopoid copepods). 为研究鲤疱疹病毒Ⅱ型(Cy HV-2)体外感染复制特征以及异育银鲫抗病毒免疫应答反应。本实验采用组织块培养法建立了异育银鲫背鳍细胞的原代培养体系。结果显示,在10 d左右可观察到组织块迁移分离出新的单层细胞,3周左右细胞可覆盖底部面积为25cm2培养瓶的底部;经Cy HV-2悬液感染离体培养的原代细胞,3 d后病毒滴度增殖至106拷贝/m L;在病毒感染6 d后出现典型的细胞病变效应;Cy HV-2感染原代细胞后,分析前期通过鱼体水平实验鉴定出的与该病毒感染相关的免疫基因:PNP5a、MPO、MHCⅠ、LYZ-C、IL-11、ITLN、PNP5a和DUSP,Real-time Rt-PCR结果显示大部分基因在细胞水平均有显著性的上调,与鱼体水平实验结果一致。本研究建立了原代培养的异育银鲫背鳍细胞,用于构建体外感染Cy HV-2病毒的细胞模型,为深入研究Cy HV-2的感染复制规律及其与宿主的相互作用关系,以及细胞水平筛选抗病毒药物实验奠定了基础。 相似文献

In this study, we identified the distribution characteristics of a mixed forest of coniferous and broad-leaved trees (a typical forest type in Hokkaido, Japan) using landscape metrics and topographic factors, and attempted to apply this knowledge to examine forest management. This approach provides a new perspective (i.e., the landscape structure) on forest management, which traditionally has been determined on the basis of individual forest stands. We first created a cover type map of the study area by means of aerial photo interpretation. The characteristics of each cover type identified from the photographs were determined using landscape metrics for each cover class. We digitized a forest administrative map (1:20,000 scale) using 20-m contours, and imported this into GIS software to produce a terrain model; on this model, we overlaid the cover types. Our examination of landscape metrics showed that most of the natural forest could be managed similarly. However, our examination of topographic characteristics revealed exceptions (e.g., areas that are difficult to regenerate) that will require particular attention when managing the natural forest. Based on the information we obtained, we proposed a guideline for sustainable forest management. From the land cover map, we proposed an “improved” cover type map to illustrate the development of a high growing stock of forest based on forest management. We compared the current cover map with the “improved” cover map and demonstrated that the improved form would have more significant effects on fauna that do not recognize differences in the proportion of the dominant species types than on those that can recognize these differences. Tree-ring-width chronology of Larix gmelinii as an indicator of changes in early summer temperature in east-central Kamchatka 总被引:2,自引:0,他引:2
We developed a 378-year tree-ring-width chronology based on 110 core samples from 55 individual trees of Larix gmelinii (Rupr.) Rupr. growing in a wide open forest close to the tree line in the Kronotsky National Park. Reflecting strong climatic
control over tree growth not only within the study area but also more extensively over central Kamchatka, our chronology was
well correlated with those from other larch sites. Response analysis with 10-day mean temperature revealed that the ring width
was primarily controlled by the temperature of early summer, i.e., of late May through late June (40 days). While the regression
models for a formal reconstruction failed to pass stringent verification tests commonly used in dendroclimatology, the relationship
between tree growth and climate was statistically significant and credible. We therefore used our chronology as a proxy of
early summer temperature. The chronology shows a cool period from the 1660s until the 1680s, followed by gradual warming until
ca. 1800, then by a slight cooling trend extending to ca. 1910, and a warming trend continuing up to the present, with decadal
fluctuations throughout the chronology. The warming trend found in our chronology over the twentieth century is generally
consistent with the ones commonly appearing in higher latitudes. 相似文献

When the influence of host species, inoculum density, temperature, leaf wetness duration, and leaf position on the incidence
of gentian brown leaf spot caused by Mycochaetophora gentianae, was examined, the fungus severely infected all seven Gentiana triflora cultivars, but failed to infect two cultivars of G. scabra and an interspecific hybrid cultivar. Inoculum density correlated closely with disease incidence, and a minimum of 102 conidia/mL was enough to cause infection. In an analysis of variance, temperature and leaf wetness duration had a significant
effect upon disease incidence, which increased with higher temperature (15–25°C) and longer duration of leaf wetness (36–72 h).
No disease developed at temperatures lower than 10°C or when leaf wetness lasted <24 h. At 48-h leaf wetness, disease incidence
was 0, 28, 77, and 85% at 10, 15, 20, and 25°C, respectively. Middle and lower leaves on the plant were more susceptible than
upper leaves. In microscopic observations of inoculated leaves, >50% of conidia germinated at temperatures >15°C after 24-h
leaf wetness. More appressoria formed at higher temperatures (15–25°C) with extended duration of leaf wetness (24–72 h). At
48-h leaf wetness, appressorium formation was 0, 8, 26, and 73% at 10, 15, 20, and 25°C, respectively. These results suggest

Genotypic variation in stomatal density and size has been reported but little is known of the genetic mechanisms behind these
leaf traits. Using 101 recombinant inbred lines derived from a cross between a tropical japonica, IR69093-41-3-2-2 and an
indica variety, IR72, we conducted a field study to determine stomatal density and size and identify quantitative trait loci
(QTL) controlling these traits under lowland conditions. Ten QTLs for stomatal density and four QTLs for size were identified
across growth stages and leaf surfaces (adaxial and abaxial). The contribution of each QTL to total phenotypic variation ranged
from 9.3 to 15.2% for stomatal density and 9.7 to 14.3% for size. The allele from IR72 increased stomatal density and that
from IR69093-41-3-2-2 increased size. The expression of the QTLs for stomatal density and size differed by growth stage indicating
that these traits might be genetically controlled depending on growth stage or that each QTL had a different function by growth
stage. Significant negative genetic correlations between stomatal density and size at both vegetative (r = −0.308**) and heading (r = −0.484**) stages were observed but no common QTL for these traits was detected across growth stages and leaf surfaces.
These results indicate that the QTLs for density and size may neither be genetically linked nor pleiotropically controlled
and findings can be used as basis for selection at the leaf level on the balance of carbon and water uptake. Further study
is needed to fully understand the mechanism underlying the observed genetic association and to elucidate the function of the

A typhoon event catastrophically destroyed a 45-year-old Japanese larch plantation in southern Hokkaido, northern Japan in September 2004, and about 90% of trees were blown down. Vegetation was measured to investigate its regeneration process and CO2 flux, or net ecosystem production (NEP), was measured in 2006–2008 using an automated chamber system to investigate the effects of typhoon disturbance on the ecosystem carbon balance. Annual maximum aboveground biomass (AGB) increased from 2.7 Mg ha−1 in 2006 to 4.0 Mg ha−1 in 2007, whereas no change occurred in annual maximum leaf area index (LAI), which was 3.7 m2 m−2 in 2006 and 3.9 m2 m−2 in 2007. Red raspberry (Rubus idaeus) had become dominant within 2 years after the typhoon disturbance, and came to account for about 60% and 50% of AGB and LAI, respectively. In comparison with CO2 fluxes measured by the eddy covariance technique in 2001–2003, for 4.5 months during the growing season, the sum of gross primary production (GPP) decreased on average by 739 gC m−2 (64%) after the disturbance, whereas ecosystem respiration (RE) decreased by 501 gC m−2 (51%). As a result, NEP decreased from 159 ± 57 gC m−2 to −80 ± 30 gC m−2, which shows that the ecosystem shifted from a carbon sink to a source. Seasonal variation in RE was strongly correlated to soil temperature. The interannual variation in the seasonal trend of RE was small. Light-saturated GPP (Pmax) decreased from 30–45 μmol m−2 s−1 to 8–12 μmol m−2 s−1 during the summer season through the disturbance because of large reduction in LAI. 相似文献

This study was conducted to determine the concordance of results for a pair of structural
isomers, 2-nitropropane (2-NP) and 1-nitropropane (1-NP), using the rat medium-term liver
carcinogenesis bioassay (Ito test) and previously published long-term carcinogenicity
tests. Male F344 rats were given a single intraperitoneal injection of DEN (200 mg/kg
b.w.) to initiate hepatocarcinogenesis. After 2 weeks, they received per os 0, 0.8, 4 or
20 mg/kg/day of 2-NP or 1-NP six times a week and were subjected to two-thirds partial
hepatectomy at week 3. Non-initiated groups receiving 0 or 20 mg/kg/day were also
included. The animals were sacrificed for quantitative analysis of GST-P-positive foci at
week 8. With the highest dose of 2-NP, significantly increased numbers and areas of
GST-P-positive foci were demonstrated as compared with the respective control but were not
noted with 1-NP. In the non-DEN-initiated groups, many small GST-P-positive foci of less
than 0.2 mm in diameter were also induced in the rats treated with 2-NP at 20 mg/kg/day
but were lacking with 1-NP. These results strongly support that 2-NP is a complete
hepatocarcinogen with a potent initiation activity, whereas 1-NP is not. 相似文献

Piceatannol is present in passion fruit (Passiflora edulis) seeds in high amounts. In this study, we isolated the second major polyphenolic compound of passion fruit seeds and identified it as scirpusin B, which is a dimer of piceatannol. We investigated the antioxidant activities and vasorelaxing effects of these polyphenols. Their antioxidant effects were measured using an in vitro 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay, and their vasorelaxant effects were determined ex vivo in rat thoracic aorta. Both polyphenolic compounds exhibited potent antioxidant activities and significant vasorelaxant effects in endothelium-intact aortas. More specifically, scirpusin B exerted a greater antioxidant activity and vasorelaxant effect compared with that of piceatannol. Additionally, the vasorelaxation effects of the compounds were induced via the NO derived from the endothelium.
